Description
To screen pH as a crystallization variable using Tacsimate one can mix together different ratios of different pH Tacsimate solutions. Refer to the pdf “Mixing Tacsimate Stocks to Screen pH” on this page. Or, one can add a buffer to the crystallizaiton reagent containing Tacsimate. Recommended final buffer concentration in the reagent well is 0.05 to 0.1 M.
Tacsimate
A pH titrated mixture of organic acids. 1.8305 M Malonic acid, 0.25 M Ammonium citrate tribasic, 0.12 M Succinic acid, 0.3 M DL-Malic acid, 0.4 M Sodium acetate trihydrate, 0.5 M Sodium formate, 0.16 M Ammonium tartrate dibasic. pH adjusted using NaOH
Measured pH of 100% Tacsimate is 4.0 at 25°C
Measured pH of 100% Tacsimate is 5.0 at 25°C

Description
Siliconized glass coverslips allow a droplet to be suspended in a position which provides near optimal conditions for vapor diffusion with the surrounding reservoir solution. These siliconized glass cover slides provide a consistent, high quality finish for crystallization experiments. The hydrophobic surface produces a drop which “stands well” and does not flatten on the glass. The siliconized surface prevents the adhesion of crystals and precipitate onto the glass surface. Use vacuum grease or oil to seal cover slide to plate. Available in 18 or 22 mm diameter circles.
HR3-254 is Greiner Bio-One 503870 siliconized glass coverslip, round, 22mm diameter, 0.19-0.22mm thick. Non-sterile. 100 pieces per inner pack. Ten inner packs, totaling 1,000 pieces per packing unit. Fit VDX Plate.

Description
SaltRx was developed by Hampton Research as a primary and secondary, salt and pH based crystallization screen for biological macromolecules.
Salt is the only primary crystallization reagent (precipitant) utilized. Based on a design of 96 conditions, the screen evaluates a broad portfolio of crystallization salts of varying concentration and pH. The selection, concentration, and pH of the salts were determined by data mining the BMCD1 and other crystallization databases, crystallization reports in the literature, and internal crystallization trials performed at Hampton Research. Based on this analysis, up to 35% of protein crystallizations involve salt as the primary crystallization reagent. SaltRx can be used as a primary crystallization screen when salt, ionic strength and pH are desired or suspected as appropriate crystallization variables. It is also useful as a secondary screen when salt only reagents/conditions from screens such as Index™, Crystal Screen™, and Grid Screen™ produce crystals and when further screening for additional salt conditions or optimization is desired. Since this screen does not contain volatile organics, it is compatible with microbatch, vapor diffusion, liquid and gel diffusion crystallization methods. SaltRx may also be used for dialysis crystallization in conjunction with Dialysis Buttons™.
The original SaltRx kit (HR2-108) contained 96 unique reagents, 10 ml each. This kit has been discontinued and converted into two 48 reagent based kits, SaltRx 1 and SaltRx 2, available separately, which when used together are identical to the original SaltRx.
SaltRx 1 contains 48 unique reagents, 10 ml each. SaltRx 1 contains reagents 1-48 of the original SaltRx 96 reagent kit.
Salt Rx 2 contains 48 unique reagents, 10 ml each. SaltRx 2 contains reagent 49-96 of the original SaltRx 96 reagent kit.
SaltRx HT contains the 96 SaltRx 1 and SaltRx 2 reagents in a single Deep Well block format.
Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity salts, polymers, organics and buffers. Individual reagents are available through the Hampton Research Custom Shop.

Stock-Options reagent kits offer precisely formulated, high quality crystallization grade reagent stocks in convenient, cost-effective kits. The chemicals utilized in StockOptions kits are the same crystallization grade, ultra-high purity chemicals utilized in the Hampton Research kits. StockOptions reagents are carefully formulated under strict quality standards to ensure reliable performance and lot-to-lot consistency.
Preformulated stocks remove calculation, measurement, and formulation errors. No more second guessing how the reagent was formulated, what specific chemical was used, when it was made, or how to precisely reproduce that reagent when it is gone and more reagent is required for additional setups.
StockOptions™ Sodium Citrate buffer kit is a preformulated, sterile filtered set of titrated buffer stocks. The StockOptions buffer stock reagents are supplied as 1.0 M stock solutions in 10 milliliter volumes. Each StockOptions Sodium Citrate buffer reagent is carefully titrated using hydrochloric acid. StockOptions Sodium Citrate is comprised of 24 unique reagents covering the pH range of 4.2 – 6.5 in 0.1 pH unit increments.
Formulated using Sodium citrate tribasic dihydrate and HCl.

Each StockOptions Sodium Citrate Buffer kit reagent is available separately as a 185 milliliter volume through the Custom Shop.

Description
The Cover Slide Vacuum Gadget™ allows one to pick up a single cover slide, invert the slide and gadget, rest the gadget on the bench so the slide rests as if on a pedestal, pipet the drop onto the slide, then use the gadget to place and release the slide onto the crystallization plate. The Gadget is a hand held vacuum bulb with a single Buna-N vacuum cup. Squeeze the bulb, press the vacuum cup to the slide, ease the squeeze on the bulb to create a vacuum and the slide is held in place at the end of the Gadget. When ready to position and place the slide give the Gadget a gentle squeeze to release the vacuum. Single bulb with 3/8″ (9.53 mm) diameter cup.

Description
Natrix, Natrix 2 and Natrix HT are based upon published reagent formulations for the crystallization of nucleic acids and protein-nucleic acid complexes. A variety of hammerhead ribozymes and other ribozymes, RNAs, DNAs, RNA-drug complexes, and RNA-protein complexes have been crystallized using the Natrix protocols.
By using sparse matrix sampling technology, The Natrix kits allow one to quickly test wide ranges of pH, salts, and precipitants using a very small sample of nucleic acid.
Natrix screens are unique in that rather than relying solely on the traditional nucleic acid precipitant MPD, Natrix screens also utilize Polyethylene glycols (PEGs) in a variety of molecular weights (200, 400, 4,000, 8,000) as well as 2-Propanol, Polyethylene glycol monomethyl ether (PEG MME), and 1,6-Hexanediol. Many of the polymeric and low molecular weight organic precipitants are combined with various monovalent salts as precipitating agents. This combination of salts and low molecular weight organics and polyalcohols, as well as the utilization of varying chain length PEGs, has proven to be a successful combination for producing nucleic acid and protein-nucleic acid complex crystals.
Natrix contains 48 unique reagents, 10 ml each and is based on the sparse matrix formulation first described by William Scott in 1995.
Natrix 2, an extension of Natrix, contains 48 unique reagents, 10 ml each. Natrix 2 is a biased sparse matrix screen based on extracting patterns from crystallization data as well as reagent formulations first described by Berger et al in 1996.
Natrix HT contains 1 ml of each reagent from Natrix and Natrix 2 in a single Deep Well block format.
Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity salts, polymers, organics and buffers. Individual reagents are available through the Hampton Research Custom Shop.

The PCT Pre-Crystallization Test is used to determine the appropriate protein concentration for crystallization screening. Sample concentration is a significant crystallization variable. Samples too concentrated can result in amorphous precipitate, while samples too dilute can result in clear drops. Precipitate and clear drops are typical crystallization screen results for reagent conditions which do not promote crystallization and are part of every crystallization screen. However, by optimizing protein concentration for the screen, the number of clear and precipitate results can often be reduced, which in turn results in more efficient sample utilization while at the same time enhancing the chances for crystallization. PCT can minimize or prevent situations where a screen results in an over abundance of precipitate or clear drops.
The PCT kit contains 4 unique, preformulated, sterile filtered reagents used to evaluate protein concentration for crystallization screening. Initially, the sample protein is mixed with two of the reagents to determine if the protein concentration is appropriate for crystallization screening. If the protein is very sensitive to salt and polymer concentration, based on initial PCT results, the protein may be evaluated using a second set of PCT reagents. PCT results will then provide insight to either the appropriate sample concentration or indicate that other diagnostic testing such as native gel electrophoresis or dynamic light scattering should be performed to demonstrate sample homogeneity appropriate for crystallization.

In carrying out animal research for our customers at Envigo we follow the principles of the 3Rs – Replacement, Reduction and Refinement. We ensure the 3Rs are considered at all levels of the company. This means we work with alternatives to animals whenever we can, we aim to carry out studies with the fewest number of animals possible, and we take measures to minimize any pain or distress before, during, and after experimental procedures.
As well as research on animals, we use a range of other approaches and methods for research such as cell cultures, in vitro techniques, computer modelling, bioinformatics, high-throughput screening and clinical trials in people. Description
The Quik Optimize reagent set is designed to deliver a flexible phosphate reagent based solubility and crystallization screening method for the crystallization of biological macromolecules. Quick Optimize allows convenient and simple reproduction of original Quik Screen conditions. The kit is especially useful for refinement and optimization of preliminary successful crystallization conditions determined using Quik Screen.
Quik Optimize is composed of two high purity, sterile filtered stock solutions, 4.0 M Sodium phosphate monobasic monohydrate (NaH2PO4 Mr 137.99 CAS Number [10049-21-5]) and 4.0 M Potassium phosphate dibasic (K2HPO4 Mr 174.18 CAS Number [7758-11-4]) formulated using ultra pure water. Final reagent pH is determined by the ratio of Sodium and Potassium phosphate reagents. The supplied dilution table makes concentration and pH determinations quick and simple. Reagent concentrations between 0.2 and 4.0 M in 0.2 M increments and pH values between 5.0 and 8.2 in 0.2 pH increments can be conveniently formulated with Quik Optimize.
The Quik Optimize formulation has been used successfully in both crystallographic and bioseparation applications for the crystallization of biological macromolecules 1-7. The phosphate reagent system in Quik Optimize is a stable, flexible, easy to reproduce, cost-effective, and simple way to screen and optimize crystallization conditions for biological macromolecules. Quik Optimize was developed by Macrocrystal Oy and is manufactured and distributed exclusively by Hampton Research.
Each Quik Optimize kit contains 4.0 M Sodium phosphate monobasic monohydrate and 4.0 M Potassium phosphate dibasic reagents, 100 ml each. These ready to use reagents are formulated using high purity salts with ultra pure water and are sterile filtered. Each kit contains a dilution table to create any pH (between 5.0 & 8.2) and any concentration (between 0.2 & 4.0 M) in 0.2 increments. Crystallization accessories are sold separately.

Description
40 mm length glass fibers for small molecule crystal mounts. Available in 0.1-0.3 mm and 0.3-0.5 mm diameter glass. Simple to use. Glue the glass fiber to a brass specimen pin. Adhesives for attaching glass fibers to the brass pins can be found in Capillary Sealants. Add a very small amount of the adhesive of choice to the tip of the glass fiber. Carefully bring the fiber into solid contact with the crystal face and leave in this orientation long enough for the glue to set firmly. Position onto goniometer head and collect data.

Description
Used during cryocrystallography to displace and reduce the amount of water (mother liquor, reagent) on the crystal after the crystal is mounted in a cryoloop. Coating the crystal with oil can minimize evaporation from the crystal and reduce exposure and slow diffusion of air (oxygen) to the crystal.
Santovac Cryo Oil offers low viscosity, incredibly low vapor pressure, low surface tension, excellent chemical inertness and excellent thermal stability.
Dip the mounted crystal into the oil to displace surface water. Or add a small amount of Santovac to the crystallization drop before mounting the crystal. After adding the Santovac, mount the crystal using a Mounted Cryoloop. Withdraw the mounted crystal from the drop and the Santovac will coat the mounted crystal. Cryogenically cool the mounted crystal.
Santovac (Santovac 5) is a five ring polyphenyl ether (PPE) with very low vapor pressure and low reactivity. The oil is highly resistant to oxidation. The only elements in the pure polyphenyl ether are oxygen, hydrogen and carbon.
The fluid is chemically stable, non-corrosive, safe and non-toxic at normal operating temperatures.
Santovac is not miscible with water, paraffin oil, silicon oil, alkanes, or Parabar 10312 (Paratone). Santovac is miscible with acetone. Shelf life 5 years.

Description
The CrystalCap ALS HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept ALS style sample mounts. The CrystalCap ALS HT attaches to a magnetic CrystalCap Vial and magnetic goniometer base. The tip of the CrystalCap accepts a Mounted CryoLoop™.
The CrystalCap Vial is a 1.8 ml cryo vial featuring two small vents. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The HR4-904 CrystalCap Vial does not have a magnet on the bottom of the vial.
The ledge free conical shape of the CrystalCap ALS HT is compatible with ALS style grippers, auto mounters and sample handlers.
The CrystalCap ALS HT features a two dimensional (2D) alphanumeric 16 x 16 data matrix code on the underside of the cap. Each cap is also color coded for CryoLoop size.

Diet induced obesity
Purified high fat diets used to induce obesity and obesity-related complications such as diabetes and metabolic syndrome typically have 40-60% of energy derived from fat. The diet tables below summarize relevant diet features for several Teklad custom research diets commonly used in rodent models.

Diets for diet-induced obesity (DIO)
Diets with 55-60% of calories from fat like TD.06414 and TD.93075 are commonly used for inducing obesity in rodents. While considered extreme compared to typical human fat consumption, these diets are effective in initiating rapid weight gain in most rodents. With higher fat content there is less room for carbohydrate, thus the carbohydrate (particularly sucrose) amount is relatively low compared to other obesity inducing diets. If you are interested in high fat and high carbohydrate, look at diets with 40-45% of calories from fat (often referred to as western diets).
As the fat level increases, pellet quality (durability) is often compromised. Some higher fat formulas are available only in non-pelleted form or require specific carbohydrate, maltodextrin, for pelleting. Depending on the fat and carbohydrate sources used, the non-pelleted form could be dense and crumbly, dough-like, or paste-like. Though a little more challenging to work with, non-pelleted diet is still used by many researchers for diet-induced obesity models as these researchers suspect the softer form may enhance obesity development.
Diets with 40-45% of calories from fat, like TD.95217 , TD.88137 , TD.06415 , and TD.08811 , represent another popular diet pattern for diet-induced obesity work. These diets have double or triple the amount of sucrose found in higher fat diets. High levels of simple carbohydrate like sucrose and fructose may help to promote hypertriglyceridemia, insulin resistance, and fatty liver. Diets with a pattern of high sucrose and high saturated or trans fat are often referred to as “Western Diets” in obesity and cardiovascular fields. Some “Western Diets” have further modifications to the fatty acid profile or even specific vitamin and minerals adjustments to be even more closely matched to a Western Diet pattern. Diets for diet-induced diabetes
Many of the same diets used for inducing obesity in rodents can be used to enhance diabetes related phenotypes like insulin resistance and glucose intolerance. However, fasting hyperglycemia characteristic of diabetes (glucose > 200 mg/dL) is uncommon with a diet only approach. Pre-feeding a high fat diet to induce a certain level of obesity and insulin resistance and then giving low-dose streptozotocin (STZ) may be an effective approach if overt hyperglycemia is desired.
Ingredient matched, low-fat DIO control diets
There are many options with different levels and types of fat in addition to different types of carbohydrate ranging from sucrose (highly refined, simple digestion) to corn starch (refined, but more complex) to resistant starch (refined, but not fully digestible). A very basic purified control diet would be AIN-93M (TD.94048 ) or AIN-93G (TD.94045 ). AIN-93 diets have a moderate amount of sucrose at ~10%, and fat is from soybean oil with a healthy fatty acid profile. Many researchers choose to compare their high fat fed animals to animals fed a natural ingredient, grain-based diet (also referred to as standard diets or chow). These diets differ in the source and level of nutrients as well as in the presence of non-nutritive factors (such as phytates or phytoestrogens). Depending on what your main comparisons are, it may be suitable to have a grain-based diet as your control/reference group. However, making such comparisons limits inferences to dietary patterns versus a specific dietary component.
