Basal mixes
You may want to add some specific ingredients in your own facility. In that case, Envigo provides basal mixes that are, by design, a starting point for additional inclusions. When you choose a basal mix diet, you simply add any ingredient(s) in a specific proportion that your require. The nutrient levels in our basal mix are adjusted to account for the dilution of the added ingredient(s), so that the final complete diet contains the desired nutrient levels.
Here are some examples of how a basal mix could be helpful to you:
- Leave all or a portion of the fat source out. The end user can add various fats and make multiple diets from one basal mix
- Make fresh diet as needed for an unstable, hazardous, or limited supply ingredient
- Add a different vitamin or mineral mix to the same basal mix to create a series of vitamin or mineral adjusted diets
If proper mixing equipment is available, and you have the capability to feed powder diet, basal mixes offer flexibility and economy.

Vitamin adjusted diets
A few vitamin adjusted formulas are shown below. Please contact us for additional formulas of this nature or for more information about altering the vitamin profile of a diet, and please read below to learn more.
Most vitamin deficient diets use vitamin-free test casein (VFT casein) as the protein source. VFT casein is casein that has been extracted with denatured alcohol to reduce the fat content from about 1% to 0.1%. In the process the content of a number of vitamins (present at low or trace levels in casein) is further reduced, making this an ideal protein source for these types of diets. Envigo produces our own VFT in-house, and we also offer this as an ingredient for those mixing their own diets.
From this vitamin deficient base, various levels of vitamins can be added back. Many formulas adjust multiple vitamins, and other nutrients.
If use of an intact protein source is not advisable, an amino acid defined diet can be used.
Contact us for other formula examples or to discuss your specific needs.

Crystal Screen • Crystal Screen 2 • Crystal Screen HT
Applications
- Primary screen for proteins, soluble peptides, nucleic acids, & water soluble small molecules
- Sparse matrix additive screen
Features
- The original sparse matrix screen
- Sparse matrix formula efficiently samples salts, polymers, organics, & pH
- Proven effective with more than 1,000 biological macromolecules
- Tube or Deep Well block format
Description
The Crystal Screen and Crystal Screen 2 reagent kits are designed to provide a highly effective and rapid screening method for the crystallization of macromolecules. The screens are simple and practical for finding initial crystallization conditions. The initial crystallization conditions for more than 1,000 proteins, peptides, oligonucleotides, and small molecules have been determined using Crystal Screen.
A highly effective approach to overcome the exhaustive search for suitable crystallization conditions is the use of a sparse matrix method of trial conditions that is biased and selected from known crystallization conditions for macromolecules. The formulation utilized in Crystal Screen and Crystal Screen 2 evaluates 96 unique mixtures of pH, salts, polymers and organics, and their ability to promote crystal growth.
Crystal Screen contains 50 unique reagents, 10 ml each and is based on the sparse matrix formulation first described by Jancarik and Kim in 1991.
Crystal Screen 2, an extension of Crystal Screen, contains 48 unique reagents, 10 ml each and is based on the formulation first described by Cudney et al in 1994.
Crystal Screen HT contains 1 ml each of reagents 1-48 from Crystal Screen and all 48 reagents from Crystal Screen 2 in a single Deep Well block format.
Ready-to-use reagents are sterile filtered and formulated with ultra-pure Type 1 water, using the highest purity salts, polymers, organics and buffers. Individual reagents are available through the Hampton Research Custom Shop.

Adjustable Crystal Mount
Applications
- Adjustable, magnetic crystal mounting with capillaries or fibers
Features
- Adjustable Z
- Magnetic mount
- Fits XYZ Goniometer Heads with Z Platform or magnetic base
- Fast, easy, and convenient crystal positioning and retrieval
Description
The Adjustable Crystal Mount is a magnetic piece (approximately 12 mm in height, 9.8 mm in diameter) designed to position a brass specimen pin and either a capillary or glass fiber onto a goniometer head for x-ray data collection.
The Adjustable Crystal Mount is engineered to fit onto the Z Platform or magnetic base within a goniometer head. The recessed bottom properly positions the crystal mount over the center of the goniometer head and prevents the mount from sliding across the goniometer head during data collection. The bottom of the crystal mount is flat and closed to prevent the brass specimen pin from protruding through the bottom of the mount. The magnetic design allows for quick, convenient, and rapid placement and retrieval of the crystal mount. Finally, the crystal mount is designed with a single allen screw which allows the Z-translation of the brass specimen pin to be adjusted (up and down) and then secured in position by tightening the screw using a goniometer head key or 0.88 mm allen key.

Description
PEGRx is a primary and secondary, polymer and pH based crystallization screen developed at Hampton Research.
It is designed to evaluate polymer based crystallization reagents and pH in low (PEGRx 1) to medium ionic strength (PEGRx 2). It is also designed for use as a secondary or optimization screen to follow the Hampton Research Index screen when polymer based reagents produce hits and interesting solubility leads.
PEGRx 1 is a crystallization reagent kit designed to evaluate an array of polymers of varying molecular weight in a low ionic strength environment versus a wide range of pH. Polymer reagents include Polyethylene glycols, Polyethylene glycol monomethyl ethers, and Jeffamines®. The molecular weight range between 200 and 20,000 is evaluated in a low ionic strength formulation. Ten different buffers are used to span the range of pH between 3.5 and 9. The primary screen variables are polymer type, polymer molecular weight, pH and low ionic strength.
PEGRx 2 is a crystallization reagent kit designed to evaluate an array of polymers of varying molecular weight in a medium ionic strength environment in the presence of additives, salts, volatile organics and polyols versus a wide range of pH. Polymer reagents include Polyethylene glycols and Polyethylene glycol monomethyl ethers. The polymer molecular weight range between 200 and 20,000 is evaluated in a medium ionic strength formulation. Ten different buffers are used to span the range of pH between 3.5 and 9. The primary screen variables are polymer type, polymer molecular weight, pH and secondary reagents which include additives, salts, volatile organics and polyols.
PEGRx 1 contains 48 unique reagents, 10 ml each.
PEGRx 2 contains 48 unique reagents, 10 ml each.
PEGRx HT contains the 96 PEGRx 1 and PEGRx 2 reagent in a single Deep Well block format.

Microbatch 72 Well Plate (Greiner)
Applications
- Microbatch crystallization
Features
- Conical, flat bottom wells
- Hydrophobic or hydrophilic versions
- Compact size
- Supplied with cover
- Terasaki plate
Description
The 72 Well Microbatch Plate consists of a 6 by 12 well layout. Paraffin, Silicon, or Al’s Oil is poured into the trough area of the plate, submerging all 72 wells under oil. Sample and reagent are then pipetted into each of the wells. The plate is available as a plasma-treated, hydrophilic version for screening or an untreated, hydrophobic version for optimization. The hydrophilic version gives better liquid handling with small volumes, while the hydrophobic version reduces crystal nucleation and helps to prevent the crystals from sticking to the plastic.
• Plate dimensions: 83.3 mm x 58.0 mm x 10.0 mm
• Conical well with 11 µl drop volume
• Flat bottom well diameter: 1.3 mm
• Supplied with cover

Seed Bead™ Kits
Applications
- Generate seeds of protein crystals
Features
- Easily generate consistent seed stocks
- Use serial dilution to control the number of seeds introduced into the drop
- Microseed matrix screening (MMS) seed stock
- PTFE, Stainless Steel, Zirconium Silicate Ceramic, or Glass Seed Bead
Description
The Seed Bead kits are used to create a seed stock for performing subsequent seeding experiments. The Seed Bead kits contains 24 Seed Bead Tubes, a special 1.5 ml microcentrifuge tube, shaped to maintain the seed in the sample during mixing. Each tube is configured with a PTFE, Stainless Steel, Zirconium Silicate Ceramic, or Glass Seed Beads.
Seeding allows one to grow crystals in the metastable zone. Crystallization in this zone provides control, reproducibility and an improved likelihood of a successful crystallization experiment. Also, crystals can grow from seeds but cannot spontaneously nucleate. By placing a seed or solution of seeds in a drop which is saturated to the metastable zone, one can use the seeds to grow larger single crystals. By controlling the number of seeds introduced into the drop, one can control the number of crystals grown. It is not practically possible to measure and know the number of seeds introduced to a drop, but by performing serial dilutions from a concentrated seed stock, one can control the number of crystals grown in the drop.
Using the Seed Bead kits, one can create crystal seed stock for subsequent seeding experiments. Crystals are placed in the microcentrifuge tube with the Seed Bead(s) and either vortexed or sonicated to generate a seed stock. Then by performing serial dilutions, one can control the number and size of crystals in the experiment.
The Seed Bead kit is useful for the preparation of seed stocks for automated and semi-automated microseeding (D’Arcy 2007, Harlos 2008).
Each Seed Bead Kit (HR2-320) contains 24 special microcentrifuge tubes, each tube containing a single 3mm PTFE Seed Bead.
Each Seed Bead Steel Kit (HR4-780) contains 24 special microcentrifuge tubes, each tube containing a six 1.6 mm Stainless Steel Seed Beads.
Each Seed Bead Ceramic Kit (HR4-781) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Zirconium Silicate Ceramic Seed Beads.
Each Seed Bead Glass Kit (HR4-782) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Glass Seed Beads.

Description
Solubility & Stability Screen is designed to assist in the identification of solution conditions which promote protein solubility and stability, and minimize protein precipitation. Solubility & Stability Screen is a solubility screen, a stability screen, and may also be used as an additive screen in the presence of a crystallization reagent.
The Hampton Research Solubility and Stability Screen can evaluate protein solubility, stability and crystallization in the presence of 94 different chemical additives sampling 17 different classes of reagents plus two controls.
Protein solubility and stability are universally required in a wide range of applications, including general biochemical studies, the preparation of proteins in pharmaceuticals, structural biology and crystallization.1-11 The preparation of a concentrated, soluble and stable protein sample can often be a difficult task as proteins often aggregate, precipitate or denature.
Thermofluor (also known as Differential Scanning Fluorimetry, DSF and Protein Thermal Shift) provides a rapid, convenient and effective means of identifying subsaturating solution conditions that may also correlate with protein heterogeneity and poor crystallization outcomes.12
Protein solubility and stability is affected by many different chemical factors including pH, buffer type, chemical additives and excipients. pH and buffer type are dominant protein solubility and stability variables and can be evaluated and optimized using the Hampton Research Slice pH kit HR2-070. Slice pH evaluates protein solubility, stability and crystallization versus 20 different buffers over the pH range 3.5-9.6. Chemical additives influencing protein solubility and stability can be evaluated using the Solubility & Stability Screen.
It is widely accepted that protein solubility and stability can be increased by the use of chemical additives.5,15 Seventeen classes of reagents are sampled by the Solubility & Stability Screen and each of these classes has been reported as important in improving sample solubility and stability.2-11
The Solubility & Stability Screen is a set of 94 high purity reagents formulated in high purity water (NCCLS/ASTM Type 1+) at 25°C and are 0.22 micron sterile filtered. The 94 solubility and stability reagents are formulated at 2 to 10 times their recommended working concentration. The remaining two reagents are water and a negative (TCA) control. A water control demonstrates the effect of diluting sample as well as sample buffer concentration. TCA, the negative control, demonstrates total sample precipitation, loss of sample solubility and loss of sample stability. The effects of the Solubility & Stability reagents can be compared with this negative control to assist in the identification of reagents promoting sample solubility and stability. 500 microliters of each reagent is supplied in a sterile 96 well polypropylene Deep Well block. The Solubility & Stability Screen reagents are compatible with the sitting or hanging drop vapor diffusion, microbatch, free interface diffusion, sandwich drop vapor diffusion, and dialysis crystallization methods utilizing water soluble reagents.

Solutions are stable at 2-8°C for months. Autoclaving is not recommended for any sulfonic acid buffer. When MES solutions are autoclaved, they turn yellow (although pH does not change measurably). The identity of the yellow breakdown product is unknown. When MES solutions are stored at room temperature, they can turn yellow within six months.
Useful pH range of MES 5.2 – 7.1
